Taxane resistance in breast cancer

چکیده

Despite recent progress in the treatment of breast cancer, disease recurrence remains a major issue to curative therapy. One of the major clinical issues is the development of drug resistance. Taxane resistance is a big challenge because taxanes, either alone or in combination, is the first-line regimens for treating breast cancer. A hierarchy of resistant mechanisms are responsible for taxane resistance. At the highest level the mechanisms involve the cancer stem cell (CSC) properties. At the middle level the mechanisms involve the multidrug resistance characters. At the lowest level the mechanisms involve a change of a specific character. Each taxaneresistant case may involve one or more levels of these resistance mechanisms. To overcome taxane resistance, future therapies need to be able to mitigate these three levels of resistance mechanisms.

Gemcitabine and vinorelbine combination chemotherapy in anthracycline- and taxane-pretreated advanced breast cancer.

PURPOSE Anthracycline and taxanes are effective agents in advanced breast cancer and prolong survival times. Some patients achieve prolongation of life with capecitabine, gemcitabine, or vinorelbine, even after failure of both anthracycline and taxanes. We analyzed the efficacy and toxicity of gemcitabine and vinorelbine combination chemotherapy in anthracycline- and taxane-pretreated advanced ...

Resistance to Taxanes in Triple-Negative Breast Cancer Associates with the Dynamics of a CD49f+ Tumor-Initiating Population

Taxanes are a mainstay of treatment for breast cancer, but resistance often develops followed by metastatic disease and mortality. Aiming to reveal the mechanisms underlying taxane resistance, we used breast cancer patient-derived orthoxenografts (PDX). Mimicking clinical behavior, triple-negative breast tumors (TNBCs) from PDX models were more sensitive to docetaxel than luminal tumors, but th...

Chemotherapy of metastatic breast cancer: what to expect in 2001 and beyond.

Chemotherapy plays an important role in the management of metastatic breast cancer. The anthracyclines (doxorubicin, epirubicin) and the taxanes (paclitaxel, docetaxel) are considered the most active agents for patients with advanced breast cancer. Traditionally, the anthracyclines have been used in combination with cyclophosphamide and 5-fluorouracil (FAC, FEC). The taxanes have single-agent a...

Taxanes for the Treatment of Metastatic Breast Cancer

Taxanes have remained a cornerstone of breast cancer treatment over the past three decades, improving the lives of patients with both early- and late-stage disease. The purpose of this review is to summarize the current role of taxanes, including an albumin-bound formulation that enhances delivery of paclitaxel to tumors, in the management of metastatic breast cancer (MBC). Since the introducti...

ression and Silencing of the Microtubule-Associated Ther tein Tau in Breast Cancer Cells

ownload microtubule-associated protein Tau has been reported to be a predictive factor for clinical response to s in metastatic breast cancer. We generated a panel of eight taxane-resistant variants from four human cancer cell lines (MCF-7, T-47D, MDA-MB-231, and BT-549). Four variants had higher levels of Tau red with their T-47D and MDA-MB-231 parental cells. Using isoform-specific primers, w...
